描述 | 新型蛋白酶 K 的基因序列来源于林伯氏白色念球菌(Tritirachium album limber)的 蛋白酶 K 基因,经过了基因定点突变和酵母细胞分泌表达、色谱纯化。经过蛋白质工程 化的新型蛋白酶 K 提高了比活性和纯度,不含有细菌内毒素,应用范围比天然蛋白酶 K 更广泛。新型蛋白酶 K 是现有蛋白酶中活性最高的品种,在 pH 4 到 pH 12 的条件下均 有活性,反应温度介于 0~75ºC 之间。在常用浓度的 SDS、尿素或 EDTA 存在时亦很 稳定。酶切割位点在脂肪族氨基酸和芳香族氨基酸的羧基端肽键,可以用于所有蛋白质 的降解。目前主要应用于基因诊断试剂盒、基因组 DNA 提取试剂盒、RNA 提取试剂盒 中去除核酸酶和其它蛋白污染。新型蛋白酶 K 的应用也包括工业领域提取非蛋白类生物 制品核酸、多糖类药物,譬如 DNA 疫苗和肝素的制备,也可用于皮肤保健去除老化角 质层细胞、糅皮、制备水解蛋白调味品等场合,取代其它蛋白酶种类。 |

使用方法 | 将蛋白酶K干粉溶解于稀释缓冲液【20mM Tris-HCl(pH 7.4), 1 mM CaCl2, 2% 甘油】中,之后用0.22 µm过滤器除菌过滤,可得20mg/ml的蛋白酶K溶液。 |
储存/保存方法 | Store at 4℃ or -20℃ for two years. |
