- Q1:用DAPI和鬼笔环肽染细胞的先后顺序是什么?
A1:一般情况下先用鬼笔环肽对细胞骨架进行染色20~40分钟,PBS清洗细胞2次,再用DAPI复染10~20分钟。可根据实际染色情况缩短或延长时间。
- Q3:DAPI染色中避光的操作到底要多严格才不至于影响结果?
A3:DAPI相对很容易染上色,避光处理时加样等操作短时见光没有关系,但是不操作的时候需要暗盒或锡纸包裹,染色后应尽快检测。
- Q2:用DAPI染细胞核时,整张图片显示蓝色是什么原因?
A2: 染色时应注意滴加量不要太多,否则会造成整张图像呈现蓝色背景,只需覆盖上薄薄的一层即可。此外,荧光染料存在淬灭的问题,建议染色后尽快检测。
